Selank’s anxiolytic and nootropic effects involve several parallel mechanisms:
GABAergic modulation: Selank enhances the activity of GABAergic interneurons and potentiates GABA-A receptor signalling without directly binding the benzodiazepine allosteric site. This produces anxiolysis without the sedation and cognitive impairment seen with benzodiazepines.
BDNF and GDNF upregulation: Selank significantly increases mRNA and protein levels of BDNF (brain-derived neurotrophic factor) and GDNF (glial cell line-derived neurotrophic factor) in hippocampal and frontal cortex regions. These neurotrophins are critical for synaptic plasticity, neuronal survival, and memory consolidation.
Locus coeruleus modulation: Reduces noradrenaline turnover in the locus coeruleus, the primary noradrenergic nucleus. Excessive LC activity underlies the hyperarousal component of anxiety - Selank’s ability to dampen this without systemic adrenergic blockade is notable.
Serotonin metabolism: Modulates serotonin uptake and metabolism in limbic structures, contributing to mood stabilisation.
