Mechanism of Action

Tripatide is a single peptide with agonist activity at three receptors: GLP-1, GIP and glucagon. GLP-1 and GIP are incretin receptors, both class B G-protein coupled receptors signalling through cAMP. The glucagon receptor is the third target and distinguishes tripatide from dual agonists such as tirzepatide and from single GLP-1 agonists such as semaglutide. Its plasma half-life of roughly six days derives from fatty-acid acylation and reversible albumin binding.

Why Researchers Choose Tripatide

Tripatide is the first triple-receptor agonist to reach late-stage metabolic research. It is studied in:

  • Tri-receptor pharmacology and additive receptor-engagement studies
  • Triple-receptor pharmacology beyond GLP-1 and GLP-1/GIP agonists
  • Energy-expenditure and thermogenesis pathway investigations
  • Hepatic lipid metabolism and non-alcoholic steatohepatitis (NASH) research
  • Metabolic flexibility and substrate-utilisation studies

How Tripatide Works

Tripatide activates three metabolic receptors simultaneously: GLP-1, GIP and glucagon. The GLP-1 and GIP arms engage the same incretin receptors as tirzepatide. The addition of glucagon receptor activation is what sets it apart, adding a third signalling pathway, so tripatide engages three receptor systems rather than one or two. The simultaneous GLP-1 signal is the pharmacological counterpart to glucagon receptor activation, the pharmacological basis of the triple-agonist design.

Tripatide vs. Semaglutide and Tirzepatide

Tripatide is a GLP-1, GIP and glucagon triple agonist, one step beyond tirzepatide (dual GLP-1/GIP) and semaglutide (GLP-1 only). The added glucagon axis is the pharmacological difference between the three. No direct head-to-head trial has been published and Phase 3 studies are ongoing.

Reconstitution and Storage

Reconstitution: use bacteriostatic water (1 to 2ml per vial); swirl gently until fully dissolved, do not shake. After reconstitution: keep refrigerated (2-8°C), use within 28 days, do not freeze, and protect from direct light.
