Tirzepatide is a single 39-amino-acid peptide engineered to activate two incretin receptors at once: GLP-1 and GIP. A fatty-acid chain enables albumin binding for a roughly five-day half-life. GLP-1 receptor activation drives glucose-dependent insulin secretion, suppresses glucagon, slows gastric emptying and reduces appetite, while added GIP receptor engagement further enhances the insulin response and influences adipose-tissue metabolism. The combined action on both pathways is what produced larger weight and glucose effects than single-receptor GLP-1 agonists in clinical trials.
