Melanotan 2 was the compound that established MC4R as the central receptor for mammalian sexual function. A 1998 Science study showed that MT-II induced spontaneous erections in MC4R-expressing rodents and that knocking out MC4R abolished the effect. That finding led directly to three developments:
- The design of PT-141 (bremelanotide) as a selective MC4R agonist
- PT-141’s FDA approval as Vyleesi for hypoactive sexual desire disorder (HSDD)
- Sustained MC4R research across obesity, sexual dysfunction and cardiovascular regulation
