Multi-Target Neurochemical Mechanism

Selank’s anxiolytic and nootropic effects involve several parallel mechanisms:

GABAergic modulation: Selank enhances the activity of GABAergic interneurons and potentiates GABA-A receptor signalling without directly binding the benzodiazepine allosteric site. This produces anxiolysis without the sedation and cognitive impairment seen with benzodiazepines.

BDNF and GDNF upregulation: Selank significantly increases mRNA and protein levels of BDNF (brain-derived neurotrophic factor) and GDNF (glial cell line-derived neurotrophic factor) in hippocampal and frontal cortex regions. These neurotrophins are critical for synaptic plasticity, neuronal survival, and memory consolidation.

Locus coeruleus modulation: Reduces noradrenaline turnover in the locus coeruleus, the primary noradrenergic nucleus. Excessive LC activity underlies the hyperarousal component of anxiety - Selank’s ability to dampen this without systemic adrenergic blockade is notable.

Serotonin metabolism: Modulates serotonin uptake and metabolism in limbic structures, contributing to mood stabilisation.

Immune modulation (tuftsin-derived): Retains the immunomodulatory properties of the parent peptide tuftsin - modulates IL-6 and IL-10 balance, potentially relevant to neuroinflammation.

Clinical Background

Selank was approved by the Russian Ministry of Health as an anxiolytic drug. Clinical trials documented:

• Significant reduction in anxiety scores (Hamilton Anxiety Scale)
• Improved cognitive performance on attention and memory tasks
• No dependence liability over the study period
• No sedation at therapeutic doses

This differentiates it meaningfully from conventional benzodiazepine anxiolytics.

Reconstitution: use bacteriostatic water; swirl gently until dissolved, do not shake. After reconstitution: keep refrigerated and use within 30 days.
